CAMP_ABEL Campylobacter venerealis IgA Antibody ELISA

Vaginal swab in PBST

COLLECTING, PACKAGING AND SHIPPING- SWAB SPECIFIC INSTRUCTIONS

  • Label the inoculated containers clearly with the Animal IDs. If you are sampling large numbers of animals, it is best to label the specimens sequentially starting at 1 and use a keylist to identify the animals.
  • Submit in an esky with ice brick/s, complying with IATA biological substances transport requirements. Samples should be packaged so they are protected from extreme heat. Samples should be maintained at 4oc while in transit
  • Schedule shipments to avoid weekend and holiday delivery of samples to the laboratory. The laboratory should receive the samples within 72 hours after collection (delivery of samples within 24 hours of collection is preferred) Include the date of collection on the submission form.
  • Please refer to the Diagnosis of Bovine Venereal Campylobacteriosis (Vibriosis) by ELISA guide for further information.

ADDITIONAL INFORMATION

In a herd situation, demonstration of antibody in vaginal mucus is the preferred method of diagnosis as it can be difficult to recover the organism from the prepuce. Collect at least ten samples from infertile heifers or cows for representative sampling of a herd.

It is best to sample when pregnancy testing reveals infertility. Testing for antibodies after calving may be unreliable, due to the extended interval from the time of infection.

Cows which have aborted can be sampled for Campylobacter ELISA from one week to three months after abortion.

Thursday and Friday

CAMP_CLT Campylobacter spp. culture

Foetal stomach contents*

C. fetus subsp. venerealis is most successfully isolated from aborted foetuses submitted to the laboratory, because the organism is not exposed to normal atmospheric conditions, and where samples can be collected without contamination and then directly cultured under microaerophilic conditions.

COLLECTING, PACKAGING & SHIPPING

  • Label all samples with the date of collection and official animal identification, making sure the key-list is also included.
  • Samples should be packaged so they are protected from extreme heat and cold and maintained at room temperature (18 – 37oc) while in transit.
  • Please submit in an esky with NO ice bricks or gel packs. Please contact the laboratory to discuss the best way to submit samples during hot weather conditions.
  • Packing should comply with IATA biological substances transport requirements.
  • Samples of ‘export animals for breeding’ should be received by the laboratory within 24 hours of sampling
  • Schedule shipments to avoid weekend and holiday delivery of samples to the laboratory. The laboratory should receive all other samples within 24 to 48 hours after collection.
  • If it is suspected that samples have been exposed to temperatures outside of the optimal range of 5oc to 37oc, target pathogen viability could have been affected. The laboratory reserves the right not to test these samples (refer to terms and conditions).

CAMP_NIP Campylobacter fetus subsp. venerealis culture

Preputial wash or scraping or vaginal swab (in CETM)

COLLECTING, PACKAGING AND SHIPPING

  • Label all samples with the date of collection and official animal identification, making sure the keylist is also included.
  • Media must be removed from the fridge and left for 30 minutes at room temperature or until it reaches ambient temperature (20-35°C) prior to use.
  • Collected samples in CETM should be stored and forwarded to the laboratory at room temperature (20-35°C) while in transit.
  • Please submit in an esky with NO frozen / cooled ice brick or gel packs, complying with IATA biological substances transport requirements. Samples should be packaged so they are protected from extreme heat and cold.
  • Schedule shipments to avoid weekend and holiday delivery of samples to the laboratory. The laboratory should receive the samples within 24 (export animals for breeding) to 48 hours after collection.
  • If it is suspected that samples have been exposed to temperatures outside of the optimal range of 20oc to 35oc, then it is likely that the target pathogen is no longer viable, and therefore the integrity of the test results are compromised. The laboratory reserves the right not to test these samples (link to T&Cs)

FMD_RTPCR Foot and Mouth (FMD) Disease Virus RT PCR*

Clotted blood (Red top)

  • 8-10ml collected in a red top tube.
  • Submit chilled
  • Be sure to avoid contamination of the container and stopper and remove all blood and faecal matter prior to packing and dispatch. Use a separate sterile needle for each animal to avoid mechanically transmitting infectious agents from one animal to another.

EDTA (Purple top)

  • 8-10ml EDTA blood collected in a purple top tube.
  • Submit chilled
  • Be sure to avoid contamination of the container and stopper and remove all blood and faecal matter prior to packing and dispatch. Use a separate sterile needle for each animal to avoid mechanically transmitting infectious agents from one animal to another.

Vesicular fluid

  • Withdraw up to 5ml of vesicular fluid with a narrow-gauge needle.
  • Place in a plain sterile specimen tube.
  • Ensure that the vial is proportionate in size to the volume of the sample to allow recovery of the sample, but total vial volume should not exceed 5mL.

Vesicular Swab

  • Swab vesicular lesions and place in phosphate buffered gelatin saline (PBGS) for transport.
  • If no PBGS is available, place swabs in 2-3 mL of sterile saline in a screw topped vial.

Swabs (oral & lesion) in PBGS

  • Vigorously swab the inside of the cheeks with sterile cotton swabs, as far into the mouth as practical and place in PBGS.
  • If it is impractical to collect swabs from the mouth, collect nasal swabs by vigorously rubbing the swab on the lining well inside the nose.

Tissue

  • Up to 2cm2 or 1g of epithelium is ideal.
  • If enough lesion is not available, collect as much as possible.
  • Samples from fresh lesions may rub off or alternatively, gently grasp the epithelium with forceps before cutting a section away.
  • Epithelium can be transported in dry sterile screw topped vials.
  • Do not mistake a large fibrin clot for epithelium - test the texture before submitting it.
  • Fibrin crumbles more easily than epithelium and fibrin will be less likely to contain virus

HIST_HE1 Histopathology – Preparation & Examination

Fixed Tissues- General

  • Fresh tissues immediately placed in formalin
  • Tissue pieces should not be greater than 1cm cubed to ensure proper fixation
    • Submit fixed in neutral buffered formalin at a 10:1 ratio of formalin: tissue

Fixed Tissues- Nervous tissues

  • Submission of whole brain is required to ensure preservation of TSE target sites
    • submit fixed in neutral buffered formalin at a ratio of 10:1 formalin: tissue
  • Brains submitted for Nervous tissues are processed once a week, separate to general tissues to ensure consistent and reliable results. Urgent cases can be processed out of cycle if clearly marked on the Submission Form however this may reduce the quality of the brain histology results, so should only be requested in extremely urgent cases.

Fresh Tissues

  • Fresh tissues can be submitted following necropsy
  • The quality of the tissues will be reduced due to the effects of autolysis so ideally fixed tissue should be submitted in neutral buffered formalin at a ratio of 10:1 formalin: tissue

Collecting, Packaging and Shipping

  • Label all samples with the date of collection and official animal identification, making sure the keylist is also included.
  • Media must be removed from the fridge and left for 30 minutes at room temperature or until it reaches ambient temperature (20-35°C) prior to use.
  • Collected samples in TFETM should be stored and forwarded to the laboratory at room temperature (20-35°C) while in transit.

If it is suspected that samples have been exposed to temperatures outside of the optimal range of 20oc to 35oc, then it is likely that the target pathogen is no longer viable, and therefore the integrity of the test results are compromised. The laboratory reserves the right not to test these samples (link to T&Cs)

Foetal Stomach Contents in TFETM or neat

  • Foetal stomach contents, collected in a sterile container, must be delivered within 6 hours of collection, otherwise it must be transported in Tritrichomonas foetus Enrichment Medium (TFEM) or neat. TFEM is available from the laboratory upon request*.
  • IMPORTANT - Do not submit chilled as organisms are sensitive to the cold.
    • Please submit in an esky with NO frozen or cooled ice bricks or gel packs.
    • Samples should be packaged so they are protected from extreme heat and cold and maintained at room temperature (18-37oC) while in transit.
    • Packing should comply with IATA biological substances transport requirements (see Additional Information).
    • Clients should schedule shipments to avoid weekend and holiday delivery of samples to the laboratory. The laboratory should receive the samples within 48 hours of collection (delivery of samples within 24 hours of collection is preferred). Include the date of collection on the submission form.
    • If it is suspected that samples have been exposed to temperatures outside of 5-37oC, target pathogen viability could have been affected. The laboratory reserves the right not to test these samples (refer to terms and conditions).

Courier services and specimen delivery

For NSW submissions, please contact the relevant Customer Service team for courier account details and to order consignment notes.

For submitters outside of NSW, Couriers can be contacted directly to make bookings and arrange collection of packages at the submitters own cost.

Commercial couriers may use either road or air transport and specimens should therefore be packed in accordance with International Air Transport Association (IATA) requirements. Most submission sent by veterinarians to the APHL for testing are defined as “Biological Substance Category B” and must be packed according to the IATA packing instructions 650 (Biological Substance Category B). As requirements for transport can change, customers should consult with their transport agent to obtain current requirements.

Samples may also be hand delivered to the site during normal business hours (8.30am - 4.30pm).
